Single-stranded DNA uses residue masses of 313.2 (A), 304.2 (T), 289.2 (C) and 329.2 (G), plus 79 for a 5' monophosphate. RNA uses 329.2 (A), 306.2 (U), 305.2 (C) and 345.2 (G), plus 159.
Double-stranded DNA adds the reverse complement as a second strand. Ambiguous bases are left out. This is an anhydrous estimate, not the mass printed on a tube of synthetic oligo, which depends on the salt form.
Why is the mass a bit different from my oligo supplier? Suppliers often report a desalted or salt-form mass, and the 5' end may be a hydroxyl rather than a phosphate. This calculator always adds the monophosphate constant so the method stays the same for every sequence.
What does pmol per microgram mean? It is how many picomoles are in one microgram of that strand. Multiply by the micrograms you have to get picomoles.
